摘 要:目的观察鲜鱼腥草挥发油减轻慢性肺损伤及降低趋化因子受体[半胱氨酸-半胱氨酸趋化因子受体2(CCR2)]高表达的作用。方法取SD大鼠,气管滴入脂多糖(LPS)诱发慢性肺损伤模型(模型组))。模型组大鼠给予不同剂量鲜鱼腥草挥发油(给药组;再分为鲜血腥草挥发油高、中、低剂量组)。各组大鼠处死后进行其支气管肺泡灌洗,取其支气管肺泡灌洗液(BALF)离心后沉淀,行WBC计数和WBC分类。取其左肺石蜡切片常规处理,行HE染色、阿尔辛蓝-过碘酸雪夫染色,分析大鼠炎症程度和杯状细胞化生。免疫组织化学染色检测其CCR2的表达。结果 1.模型组大鼠BALF中WBC总数和中性粒细胞数明显高于正常组,肺组织呈不同程度、不同数量的支气管、细支气管慢性炎症。2.经不同剂量鲜鱼腥草挥发油灌胃后能减少WBC总数及中性粒细胞数,降低炎症程度;尤其是鲜鱼腥草挥发油高剂量组。3.给药组大鼠肺泡内CCR2阳性染色区域减少,染色程度减轻;通过图像分析,给药组大鼠肺组织中CCR2的表达较模型组显著降低。结论鲜鱼腥草挥发油可减轻LPS引起的大鼠慢性肺损伤,并能抑制肺泡内CCR2的高表达。Objective To observe the effect of the volatile oil from fresh herba houttuyniae on relieving and inhibiting the chronic lung injury and reducing the high level of cysteine-cysteine chemokine receptor 2(CCR2) expression in lung tissue of rats.Methods Chronic lung injury model was induced by intratracheal instillation of lipopolysaccharide(LPS)(model group).The model rats were treated with a series doses of the volatile oil from fresh herba houttuyniae(including low,middle,high dosage groups).The lung of the killed rats was lavaged with saline,and the bronchoalveolar lavage fluid(BALF) was centrifuged,the total numbel of leukocyte and differential leukocyte numbers of smear sedimentationwhich were counted.Pathological sections were made in left lung.The change of airway inflammation and goblet cell pro-liferation were investigated by HE and alcian blue-periodic acid schiff staining.Immunohistochemistry section were made in left lung and the effect of volatile oil from fresh herba houttuyniae on the lever of CCR2 expression was further observed.Results 1.Compared with normal group,leucocytes and neutrophile number in bronchoalveolar lavage fluid of model group were significantly increased,different levels of chro-nic inflammation in bronchi and bronchioles were observed.2.Leucocytes and neutrophile number and inflammtion severity were significantly reduced after administration of the volatile oil from fresh herba houttuyniae,especially in high dosage group.3.Stained CCR2 area on alveolus was faint in color and small in size,by analyzing the gray lever of CCR2 with image-analyzed system,the lever of CCR2 expression were lo-wer than that in model group.Conclusion The volatile oil from fresh herba houttuyniae can relieve and inhibit the development of chronic lung injury by LPS rats,and reduce the level of CCR2 expression in lung tissue.
