摘 要:羧基荧光素乙酰乙酸(CFSE)标记细胞与流式细胞仪相结合的技术在免疫学领域应用越来越广泛。该技术已经由淋巴细胞扩展到应用于肿瘤细胞、干细胞、自然杀伤细胞(NK细胞)甚至是细菌的研究,由检测细胞增殖拓展到体内示踪等研究。此外,对于该项技术的热点还集中在对其数据的分析、与传统方法的比较以及标记的方法的研究。尤其是对细胞增殖的结果的定量分析已经成为生物数学领域的一个热点,也为机制研究提供了更丰富的信息。羧基荧光素乙酰乙酸具有动态检测细胞增殖动力学,在单细胞水平上供信息等优势,有替代传统方法(如^3H掺入法、MTT法等)的趋势。It is increasingly wide that the application of the technique that combines the carboxyfluorescein diacetate succinimidyl ester(CFSE) with the flow cytometer.This technique has been extended from lymphocyte research to many other cells research, including the tumor cell,stem cell,natural cell and even the bacteria,from cell proliferation detection to cell tracing in vivo.Moreover, researchers also pay their attention on the data analysis,the comparison with the traditional methods,and the staining process of the technique,especially the data analysis,which provides information for the explanation of cell proliferation mechanism.CFSE is able to monitor the proliferation of cell dynamically and provides information in the level of the single cell,thus has the trend to substitute the traditional methods,such as ~3H-thymidine incorporation test and the MTT test.
