胃宁方对胃癌前病变大鼠转化生长因子β/Smads信号通路的影响

摘　　要：[目的]探讨胃宁方对胃癌前病变(PLGC)大鼠转化生长因子β(TGF-β)/Smad信号通路的影响。[方法]60只大鼠随机分为6组:空白对照(空白)组,模型组,维甲酸组,胃宁方低、中、高剂量(低、中、高剂量)组,每组10只,采用以N-甲-N-硝基-亚硝基胍(MNNG)为主的4因素联合造模法建立PLGC大鼠模型。维甲酸组给予维甲酸水溶剂1.0 g/kg;低、中、高剂量组分别给予胃宁方生药浓度为1.0、2.0、3.0 g/ml的煎剂,10 ml/kg,均每日1次灌胃。12周后观察各组大鼠一般情况、胃黏膜病理组织学变化,以及对TGF-β1、Smad4 mRNA表达和细胞外信号调节激酶(ERK1)水平的影响。[结果]胃宁方各剂量组大鼠活动、饮食等一般情况好转;模型组与空白组比较,TGF-β1、ERK1表达水平明显增高,Smad4表达水平明显降低;低、中、高剂量组与模型组比较,TGF-β1、ERK1表达水平下调,Smad4表达上调,差异均有统计学意义(P<0.05,<0.01)。[结论]胃宁方可改善PLGC大鼠胃黏膜病理组织状况,下调TGF-β1mRNA表达及ERK1水平,上调Smad4 mRNA表达。Objective]To study the effect of Weining decoction on the TGF-β/Smad signal transduction pathway in rats with precancerous lesions of gastric carcinoma（PLGC）.[Methods]Sixty rats were randomly divided into six groups,including blank group,model group,retinoicacid group,Weining decoction low-dose,middle-dose and high-dose group,n=10 in each.The rat model of PLGC established by four pathogenic factor in which MNNG was mainly used.Retinoicacid was given 1.0g/kg per day in retinoicacid group;Weining decoction of 1.0,2.0,3.0 g/ml drug concentration were given 10 ml/kg per day in low-dose,middle-dose and high-dose group.After 12 weeks,the general conditions,the pathological changes in rat gastric mucosa as well as TGF-β1,Smad4 mRNA expression and EPK1 levels were observed.[Results]General conditions,such as activity and eating were improved in all Weining decoction groups;Compared with the blank group,the expressions of TGF-β1 and ERK1 in the model group were significantly increased,that of Smad4 was significantly reduced,the differences between these two groups was statistically significant（P〈0.05,〈0.01）;Compared with that of model group,the expressions of TGF-β1 and ERK1 in different dose of Weining decoction group were significantly reduced,Smad4 was signifieantly increased,the differences was statistically significant（P〈0.05,〈0.01）.[Conclusion]Weining decoction can improve the pathological conditions of gastric mucosa in rat with PLGC;And it can also prevent the progress of PLGC by reducing TGF-β1 mRNA expression and ERK1 levels,increasing the expression of Smad4 mRNA.
