摘　　要：目的研究Wnt3基因在骨髓间充质干细胞增殖过程中的变化和作用。方法4贴壁培养法分离大鼠骨髓间充质干细胞，将传至第五代细胞接种于培养皿，进行细胞计数，描绘生长曲线，并于第1，3，5天取细胞进行实时定量PCR检测Wnt3表达情况。结果与空白组相比，第五代骨髓间充质干细胞于第1，3，5天表达增加（P＜0.05）。结论Wnt3在骨髓间充质干细胞增殖过程中表达增加，提示在此过程中，经典Wnt信号通路被激活。Objective To investigate the alteration and role of Wnt3 in the process of BM--MSCs proliferation. Methods Rat bone marrow mesenchymal stem cells were isolated from the bone marrow of SD rat and cultured using adherence method. P5 （Fifth passage） cells were cultured in Six--well plates, the cell number was counted and the growth curve was made. Total RNA was isolated from cells inclu- ding day 1, day 3 and day 5, the expression of Wnt3 gene was determined by real--time quantitative PCR assay. Results Compared with the blank group, the expression of Wnt3 of day 1, day 3 and day 5 increased （P〈 0.05）. Conclusions In the period of proliferation of BM-- MSCs, the expression of Wnt3 gene increased, suggesting that the canonical Wnt singal way was activated.
