摘  要:目的:探讨牛珀至宝丹对老年记忆减退大鼠学习记忆能力及脑TGF-β1表达和NPCs增殖的影响。方法:以成年大鼠平均逃避潜伏期的均值加两倍标准差和加一倍标准差为参照标准,将老年大鼠随机分为记忆正常组、记忆减退组、牛珀小剂量组及牛珀大剂量组。牛珀至宝丹0.05g/kg、0.1g/kg连续给药15d后,BrdU标记处于增殖状态的NPCs,Morris水迷宫进行学习记忆能力检测,免疫组化检测鼠脑尾壳核、皮质区TGF-β1表达及SVZ、DG部位BrdU阳性细胞数。结果:定位航行实验第4~5d,牛珀大剂量组大鼠平均潜伏期明显缩短,与记忆减退组比较,有显著性差异(P<0.05);空间探索实验,牛珀大剂量组大鼠第Ⅱ象限停留时间、第一次平台穿越时间、穿越平台次数与记忆减退组比较,有显著性差异(P<0.05);牛珀大剂量组大鼠脑尾壳核及皮质区TGF-β1蛋白表达明显增多,SVZ及DG部位BrdU阳性细胞数明显增加,与记忆减退组比较,有显著性差异(P<0.05)。结论:牛珀至宝丹对老年记忆减退大鼠学习记忆能力有明显促进作用,能显著促进内源性NPCs的增殖,其机理可能与牛珀至宝丹增强TGF-β1蛋白在大鼠脑内的表达有关。Objective:To discuss the influence of NPZBD on abilities of learning and memory,expression of TGF-β1 protein and proliferation of NPCs in aged hypomnesia rats.Methods:The aged rats were randomly divided into normal memory group,hypomnesia group,NPZBD low dose treatment group and NPZBD large dose treatment group with by reference standard of an average of the mean escape latency add twice the standard deviation and standard deviation of adult rats.The aged rats of treatment group were administrated with NPZBD 0.05g/kg/d and 0.1g/kg/d for 15days.BrdU was injected into the different aged rats to label the proliferating NPCs.ALL rats were tested abilities of learning and memory with Morris water maze after the administration and detected the changes of TGF-β1 in zone of Caudate putamen and cortex and BrdU in zone of SVZ and DG with immunohistochemistry.Result:In 4th to 5th day of splace navigation,the escape latenc of rats in NPZBD large dose treatment group were significantly shorter than hypomnesia group,there is significant difference(P〈0.05);In spacial probe test,the time in second quadrant,time of first through platform,times of through platform of rats in NPZBD large dose treatment group was significant different with the rats in hypomnesia group(P〈0.05);The expression of TGF-β1 protein in zone of Caudate putamen and cortex and BrdU in zone of SVZ and DG of aged rats in NPZBD large dose treatment group was significant different with the rats in hypomnesia group(P〈0.05).Conclusion:NPZBD had obvious promotion on ability of learning and memory in aged hypomnesia rats and enhanced proliferation of endogenous NPCs.The mechanism might be concerned with NPZBD enhanced expression of TGF-β1 protein in brain.
