六月青总皂苷抗鸭乙型肝炎病毒作用研究

摘　　要：目的:观察六月青总皂苷(the terpenoids of Liuyueqing,TLYQ)体内抗鸭乙型肝炎病毒(duck hepatitis B virus,DHBV)的作用。方法:将DHBV DNA阳性麻鸭随机分为六月青总皂苷大、中、小剂量治疗组、拉米夫定组和模型组,分别给予相应药物进行干预。用药前、用药第7、14d及停药第7d,取静脉血用实时荧光定量PCR法检测血清DHBV DNA含量。治疗后取肝脏组织作病理学检查。结果:拉米夫定组用药后血清DHBV DNA水平迅速降低,停药后立即反跳。六月青总皂苷大剂量组第7d、14d即能明显抑制DHBV DNA,停药后仍有一定的抑制作用。中、小剂量组与模型组比较有一定的病毒抑制作用。病理改变各组在统计学上无明显差异。结论:六月青总皂苷可有效地抑制DHBV DNA,其作用有明显的量效和时效反应关系。Objective：To observe the inhibitory effect of the terpenoids of Liuyueqing（TLYQ）on duck hepatitis B virus in vivo.Methods：Ducks with congenital infection of DHBV were randomly divided into five groups：high,medium,low-dose TLYQ-treated group,lamivudine-treated group and untreated group.The ducks in the lamivudine-treated group were fed lamivudine with a dose of 50mg/kg once.Ducks in the three-dose TLYQ-treated groups were treated with different doses of TLYQ for 14 days respectively.The serum content of DHBV DNA was determined by FQ-PCR method before and on the 7th,14th day of the treatment,and the 7th day after the treatment.The histopathological changes of liver were evaluated after the treatment.Results：Lamivudine showed a rapid inhibiting effect on DHBV DNA,but this effect didn＇t last long,and the serum level of DHBV DNA increased again after treatment.The serum level of DHBV DNA dropped markedly in the high-dose TLYQ-treated group on the 7st and 14st day.Low-dose and medium-dose TLYQ also showed certain inhibiting effect on DHBV DNA.The features of liver histopathological changes in TLYQ treated groups showed no statistical significance as compared with that in the untreated group.Conclusions：TLYQ can inhibit significantly DHBV DNA,and its inhibitory effects were dose-and time-dependents.
