大鼠经颅高压电烧伤后CD11b/CD18表达对白细胞黏附的影响及乌司他丁干预作用  被引量:5

摘  要:目的观察经颅高压电烧伤后大鼠CD11b/CD18的表达对白细胞黏附的影响,探讨乌司他丁对CD11b/CD18表达和白细胞黏附的干预作用。方法将144只大鼠分为对照组、电伤组和干预组,每组48只。用TC-30-20kVA型调压器和YDJ-10kVA型实验变压器复制大鼠经颅高压电烧伤动物模型,用Epics-XLⅡ型流式细胞仪检测3组伤前15min、伤后5min、1h、2h、4h、8h大鼠白细胞CD11b/CD18的变化,用BVPM-15型布莱德多项投影显微镜系统观测以上各时相肠系膜微静脉白细胞黏附数,用LISCA激光多普勒微循环图像仪检测以上各时相胸部皮肤微循环灌流量。结果电伤组和干预组伤后各时相CD11b/CD18表达量高于伤前15min,也高于对照组同时相,两组CD11b/CD18表达量于伤后5min达高峰,之后呈下降趋势;干预组伤后各时相的CD11b/CD18的表达量均低于电伤组各相同时相,但高于对照组各相同时相。两组白细胞黏附数及微循环灌流量与CD11b/CD18有相似的变化趋势。结论经颅高压电烧伤导致大鼠白细胞CD11b/CD18表达增强、白细胞黏附数增加及微循环灌流量下降,乌司他丁可降低白细胞CD11b/CD18表达、减少白细胞黏附及增加微循环灌流量。Objective To explore the influence of expression of the rat CD11b/CD18 on leukocyte adhesion and the therapeutic effects of ulinastatin after high-voltage electricity burns(HEB).Methods 144 rats were randomly divided into three groups of 48 rats each:control group,electrical burns group and treatment group.The system made up of a TC-30-20 kV A type voltage regulator and YDJ-10 kV A type transformer was used to reproduce the HEB animal model.The Epics-XLⅡflow cytometer was used to detect CD11b/CD18 in leukocytes 15 minutes before HEB and 5 minutes,1,2,4 and 8 hours after HEB.The Bradford Variable Projection Microscope(BVPM-15)was used to measure the leukocytes adhering to mesenteric venule and the LISCA Laser Doppler Hemoperfusion Image instrument(LDPI)was used to measure perfusion of chest skin microcirculation.Results Compared with 15 minutes before HEB,the expression levels of CD11b/CD18 increased in electrical burns group and treatment group after HEB,which peaked 5 minutes after HEB and was higher compared with control group.Compared with the electrical burns group in each time phase,the expression levels were decreased in the treatment group,but higher than in the control group.The perfusion,the leukocytes adhesion and expression levels of CD11b/CD18 changed in the same way.Conclusion High-voltage electricity can lead to potentiation of the expression of CD11b/CD18 in leukocytes of rats,increase the leukocytes adhesion,and decrease the perfusion.Ulinastatin can decrease the expression of CD11b/CD18 and the leukocytes adhesion,and increase the microcirculation perfusion.
