摘 要:以美女樱品种"Quartz"离体叶片为外植体,探讨了不同细胞分裂素、暗培养、硝酸银质量浓度以及不同植物生长调节剂组合对不定芽再生的影响。结果表明:6-BA的诱导效果好于TDZ。在培养基MS+6-BA4.0mg/L+IBA0.15mg/L+蔗糖30g/L上可以使美女樱叶片不定芽的再生率达26.8%。2mg/LAgNO3可以将美女樱叶片的不定芽再生率提高到31.3%,并缩短不定芽发生时间。暗培养5d可以促进美女樱叶片不定芽的再生,活性炭对美女樱叶片离体再生有一定抑制作用。The ideal regeneration system of Verbena hybrida Quartz by leaf explants was established.In this study,the effects of plant growth regulators,dark periods and AgNO3 concentrations on shoot regeneration from leaves of verbena were investigated.The results showed that the regeneration rate of 26.8%,was obtained on the medium MS supplemented with 4.0 mg/L 6-BA and 0.15 mg/L IBA and 30 g/L sucrose.6-BA was more effective than TDZ in inducing shoot regeneration from leaves of Verbena.2 mg/L AgNO3 could increase the shoot regeneration rate to 31.3% and shorten regeneration time.5 days of dark treatment could promote shoot regeneration while charcoal inhibited shoot regeneration.
