摘  要:目的应用组织芯片技术,在免疫组化超敏二步法中对二抗回收的可行性进行探讨。方法根据所测试抗体的不同,选取本院病理科48例中性甲醛固定、石蜡包埋的组织标本(包括正常及病变的组织),用组织芯片制作仪构建1个组织芯片蜡块。用免疫组化超敏二步法试剂分别测试3种单克隆抗体(Vimentin、CK和Ki-67)。结果经平行的免疫组化染色后,在48例的试验中,未经使用的二抗与经一次使用后回收的二抗显色的结果相同或相似。结论在此次免疫组化染色测试中,组织芯片显示了其信息含量多、可比性强及省时、省力等优点,二抗显色的结果相同或相似(P>0.05),提示超敏二步法中二抗回收是可行的。Objective To explore the feasibility of recovery of secondary antibody in immunohitochemical staining by using tissue microarray technique.Methods According to different testing antibodies,48 neutral formaldehyde fixed and paraffin-embedded samples were chosen to test used secondary antibody in tissue microarray,each of which would be used to test the monoclonal antibody such as Vimentin,CK or Ki-67 by HRP detection system.Results The effect of staining with both unused secondary antibody and used secondary antibody recovered once were similar.Conclusion Secondary antibody which recovered from immunohistochemical staining is feasibility.
