摘  要:目的:研究皱瘤海鞘醇提物体外抗单纯疱疹病毒2型(HSV-2)的效果及其作用机制。方法:用HSV-2在Vero细胞上的增殖为模型,运用细胞病变法(CPE法)和四甲基偶氮唑盐比色法(MTT比色法)对皱瘤海鞘醇提物体外抗HSV-2的药效学作用进行评价,在HSV-2复制的不同阶段用药物进行干预,探讨皱瘤海鞘醇提物对HSV-2复制的影响机制。结果:皱瘤海鞘醇提物的毒性低,对Vero细胞的CC50值大于10mg.mL-1。对HSV-2有良好的抑制作用,EC50值为0.40mg.mL-1,TI>25。皱瘤海鞘醇提物对HSV-2具有杀伤作用和抑制HSV-2复制,对HSV-2的吸附作用影响较小。结论:皱瘤海鞘醇提物对HSV-2复制具有良好的抑制作用。ORJECTIVE To study the role of ethanol extract of Styela plicata on inhibiting herpes simplex virus 2(HSV-2) and its mechanism of action in vitro. METHODS The model that HSV-2 proliferating in the Vero cells was applied, cytopathic effect assay and MTT assay was applied to evaluate the effect of water extract of Styela plicata on HSV-2 in vitro. Ethanol extract of Styela plicata in various concentrations was applied to different stages of HSV-2 replication cycle, the mechanism that ethanol extract of Styela plieata inhibiting HSV-2 was investigated. RESULTS Toxicity of ethanol extract of Styela pticata was very low, the CC50 value of ethanol extract of Styela plieata was greater than 10 mg.mL-1 , EC50, value was 0. 40 mg.mL -1 , TI〉25. Alcohol extract of Styela plicata had a strong role of killing HSV-2, and could strongly inhibit HSV-2 replication, had no obvious effect on the adsorption of HSV-2. CONCLUSION Ethanol extract of Styela plicata has a good role of inhibiting HSV-2 infection in vitro.
