摘  要:目的:探讨生长分化因子-15(GDF15)在心脏成纤维细胞中表达及意义。方法:分离原代大鼠成纤维细胞,分为对照组,ET-1刺激组(10^(-6)mol/L),GDF15刺激组(3 pmol/L);利用realtime-PCR法检测GDF15 mRNA表达,用Western印迹法、ELISA法检测GDF15蛋白表达;用MTT和流式细胞术检测GDF15对成纤维细胞增殖的影响。结果:心脏成纤维细胞生理条件下表达GDF15,ET-1诱导GDF15表达增加,ET-1组GDF 15 mRNA是对照组(1.20±0.03)倍(P=0.02),上清中分泌的蛋白则是(3.27±0.81)pg/mL,高于对照组(0.41±0.17)pg/mL (P=0.013)。外源性给予GDF15刺激后,MTT测得OD值与对照组比较为(0.62±0.03)比(0.48±0.03),(P=0.029);而流式细胞术检测S期比例较对照组升高近3倍(9.62±1.17)%比(3.80±0.78)%,P=0.008),均提示能够促进成纤维细胞增殖。结论:GDF15在成纤维细胞中有表达并具有促进成纤维细胞增殖作用,提示其可能参与心肌纤维化的病理过程。Objective:To investigate the expression of Growth differentiation factor-15(GDFlS) and the effect of GDFI5 in the proliferation of fibroblasts.Methods:Primary myocardial fibroblasts were isolated from neonatal rats and divided into three groups treated with different conditions: ET-1(10^-6 mol/L),GDF 15(3 pmol/L) and nothing.The expression of GDF 15 mRNA was detected by realtime-PCR and the expression of GDFI5 protein was shown by Western-Blot and Elisa.The proliferation of fibroblast induced by GDF-15 was measured by MTT and Flow cytometry. Results:GDF15 is upregulated in ET-1 group:the GDF15 mRNA is 1.20±0.03 fold of control group (P=0.02), and protein in the supematant is (3.27±0.81)pg/mL vs (0.41±0.17)pg/mL(P=0.013).GDF15 could enhance the MTT OD value from 0.48±0.03 to 0,62±0.03 (P=0.029),it could raise the cell in S phase nearly 3 times more than control group (9.62±1.17)% vs (3.80±0.78)%,P=0.008). Conclusinn:GDF15 is uprcgulatvd by ET-1 in myocardial fibroblasts.and it could enhance the proliferation of fibroblast,which may participate the progression of myocardial fbrosis.
