摘  要:目的:研究肝纤维化形成过程中RECK基因的表达与基质金属蛋白酶-2(MMP-2)活性之间的相关性。方法:SD雄性成年大鼠(25只)随机分为对照组(5只)和肝纤维化组(20只)。对照组每周2次纯花生油皮下注射,剂量为3ml·kg-1,第8周全部处死。肝纤维化组每周2次用50%CCl4油溶液皮下注射,剂量为3ml·kg-1,建立起肝纤维化模型,并分别于第1、2、4、6、8周末各处死4只动物,留取肝组织备用。天狼猩红染色判断肝纤维化程度,RT-PCR法检测RECK基因的表达情况,明胶酶谱法检测肝组织MMP-2的活性。结果:MMP-2的活性随肝纤维化的形成而逐渐升高,而RECK基因表达的变化与之相反,RECK的表达和MMP-2的活性呈负相关(P<0.05)。结论:在大鼠肝纤维化形成过程中,RECK的表达与MMP-2活性可能有密切关系,前者对后者可能起抑制作用。Objective:To investigate the relationship between RECK expression and matrix metalloproteinase-2(MMP-2) activity in the formation process of hepatic fibrosis in rats.Methods:Spraque Dawley male rats(25 rats) were randomly divided into two groups:control group(5 rats) and hepatic fibrosis group(20 rats).The control group were injected hypodermically by oil solution twice a week(the dosage is 3 ml·kg-1) and all of them were sacrificed at the end of eighth week.The hepatic fibrosis model was induced by 50 % CCl4 in oil solution twice a week(the dosage is 3 ml·kg-1) in the experiment group and four rats were randomly sacrificed at the end of the first,second,fourth,sixth,eighth week respectively.The collagen deposition,RECK expression and MMP-2 activity in the liver tissues were detected by Sirius red staining,RT-PCR and zymography respectively.Results:It was found that MMP-2 activity increased gradually in the formation process of the hepatic fibrosis.But RECK expression had reversed change at the same time.There was negative correlation between RECK expression and MMP-2 activity in the liver tissues(P0.05).Conclusions:The results suggest that MMP-2 activity and RECK expression may be closely related in the formation process of hepatic fibrosis.RECK maybe have depressant effect on MMP-2 activity.
