钙调素拮抗剂EBB抑制ES-2细胞转移作用机制初探

摘　　要：目的研究钙调素拮抗剂EBB-O-(4-乙氧基-丁基)-小檗胺体外对人卵巢癌ES-2细胞的转移抑制作用及其机制。方法采用MTT法测定EBB对ES-2细胞的增殖抑制作用;Transwell小室法分析细胞侵袭能力,细胞损伤实验检测细胞迁移能力;共聚焦显微镜技术检测细胞内[Ca2+]i变化。结果EBB对ES-2细胞具有增殖抑制作用,IC50为(13.67±1.56)μmol.L-1,EBB使ES-2细胞的侵袭迁移能力明显下降(P<0.01),伴随[Ca2+]i呈时间及剂量依赖性增高。结论钙调素拮抗剂EBB可抑制ES-2细胞增殖,降低ES-2细胞的侵袭转移能力,与细胞内游离钙离子浓度增高相关。Aim To investigate the potential suppressing effect of calmodulin antagonist EBB on metastasis-associated properties of human metastatic ovarian clear cell adenocarcinoma cell ES-2.Methods MTT assay was used to assess the growth inhibition of EBB on ES-2 cells.The invasive capacity and motility potential were determined by Transwell chamber assay and Wound assay.[Ca^2＋]i was observed under the laser scanning confocal microscopy.Results EBB inhibited the proliferation of ES-2 cells in vitro.The IC50 on ES-2 cells was（13.67±1.56）μmol·L^-1.The invasive ability and motility potential of ES-2 cells were decreased after exposure to 3,7 and 14 μmol·L^-1 EBB respectively（P〈0.01）.EBB enhanced the [Ca^2＋]i in a dose and time dependent manner.Conclusions Calmodulin antagonist EBB is effective in suppressing ES-2 cell metastasis-associated properties.[Ca^2＋]i is related with the process.The results suggest that EBB and other calmodulin antagonist might be valuable candidate drugs to inhibit invasion of tumors.
