摘 要:目的研究植物雌激素α-玉米赤霉醇对去卵巢大鼠骨生物力学的影响。方法 40只6月龄雌性SD大鼠随机分为4组:假手术组(Sham)、模型组(OVX)、雌二醇组(OVX+E2)、α-ZAL组(OVX+α-ZAL)。术后三周假手术组和模型组灌胃给予生理盐水10mL/kg·d,雌二醇组给予戊酸雌二醇1mg/kg·d,α-ZAL组给予α-ZAL1mg/kg·d。12周后处死动物,分离右侧股骨,测定股骨最大载荷、最大挠度、弹性载荷、弹性挠度、最大应力和最大应变。结果α-ZAL组大鼠股骨最大载荷、最大挠度、弹性载荷、弹性挠度、最大应力和最大应变等骨生物力学指标较模型组显著提高。结论α-玉米赤霉醇能改善去卵巢大鼠骨生物力学性能,提示其具有良好的防治绝经后骨质疏松症的应用前景。Objective To examine the effects of the phytoestrogen α-zearalanol (α-ZAL)on biomechanics of bone in an animal model of established osteoporosis. Methods 40 six-month old SD female rats were randomly divided into four groups: Sham group,model group (ovariectomized),estrogen group (ovariectomized and fed estrogen), α-ZAL group (ovariectomized and α-ZAL). Three weeks post-surgery, the sham group and model group were treated daily by oral gavage with physiological saliner10 mL(/kg·d), and the other two groups were fed with estradiol valerate 1 mg(/kg·d)or α-ZAL 1 mg(/kg·d)for 12 weeks. After treatment, maximum load, maximum deflection, elastic load, elastic deformation, maximum stress and maximum strain of right femurs were observed. Results After treatment, the maximum load, maximum deflection, elastic load, elastic deformation, maximum stress and maximum strain of right femurs in α-ZAL group were significantly increased than that of model group. Conclusion α-ZAL can improve biomechanical properties of bone in ovariectomized rats, having good application prospects in prevention and cure of postmenopausal osteoporosis.
