摘 要:目的合成莪术醇全抗原并加以鉴定。方法先用莪术醇与琥珀酸酐为原料,以三氯甲烷为溶剂,4-二甲基氨基吡啶(DMAP)作催化剂,反应合成半抗原莪术醇琥珀酸单酯,然后,用1-乙基-3(3-二甲基氨基丙基)碳二亚胺法将莪术醇琥珀酸单酯偶联到牛血清白蛋白,合成了莪术醇的人工抗原,并用激光解析飞行质谱(MALDI-TOF-MS)进行了鉴定,利用质谱特征求得了偶联物中莪术醇与牛血清白蛋白的量,计算出结合比。结果成功地合成了莪术醇人工抗原,其结合比为19.6。结论用1-乙基-3(3-二甲基氨基丙基)碳二亚胺偶联法可合成莪术醇的完全抗原。Objective To synthesize and identify the artificial antigen of curcumol.Methods Firstly,a hapten of curcumol,curcumol-hemisucinate(Cur.-HS) was synthesized using material of curcumol to react with succinic anhydride in chloroform and trace 4-dimethylaminopyridine(DMAP) as catalyst.Then the hapten was conjugated to bovine serum albumin(BSA) using 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide,(EDC) method and the characterization of the antigen was examined by MALDI-TOF-MS,and the conjugation ratio of curcumol to BSA was determined with MALDI-TOF-MS.Results The artificial antigen was successfully synthesized and identified by MALDI-TOF-MS.The conjugation ratio of curcumol to BSA was 19.6.Conclusion The artificial antigen of curcumol is successfully synthesized by EDC method.
