摘 要:目的用高效液相色谱法比较蒙药材草乌及其炮制品中乌头碱、次乌头碱、新乌头碱的含量。方法色谱柱为Dia-monsil C18柱(250mm×4.6mm,5μm);流动相A为乙腈-四氢呋喃(25:15,V/V),B为0.1mol.L-1醋酸铵-冰醋酸(100:0.05,V/V),梯度洗脱;检测波长为235nm;流速1.0ml.min-1;柱温为30℃;进样量10μl。结果蒙药材草乌经特殊炮制后,炮制品中乌头碱、次乌头碱、新乌头碱的含量均低于草乌生品,诃子制草乌中含量最低,甘草制草乌含量次之,酸奶制草乌含量最高。结论 3种炮制方法使草乌药材中双酯型生物碱含量大大降低,可用于指导蒙医临床用药。Objective To compare the contents of aconitine,hypaconitine and mesaconitine in Radix Aconiti Kusnezoffii and its mongolian distinct preparations by HPLC.Methods Diamonsil C18(250 mm×4.6 mm,5 μm) was used as chromatographic column,acetonitrile-tetrahydrofuran(25:15,V/V) and 0.1 mol·L^-1 ammonium acetate-acetic acid (100:0.05,V/V) in gradient elution mode as mobile phase.The flow rate was set at 1.0 ml·min-1,the detection wavelength at 235 nm and the column temperature was at 30℃.Results After Radix Aconiti Kusnezoffii was processed by Mongolian distinct technology,the contents of aconitine,hypaconine and mesaconitine in its preparations were less than those in unprocessed Radix Aconiti.Kusnezoffii,and in these preparations the total content of three alkaloids thin myrobalan preparation of Radix Aconiti Kusnezoffii was minimum,that in liquorice preparation of Radix Aconiti Kusnezoffii was second,and that in acidophilous milk preparation of Radix Aconiti Kusnezoffii was maximum.Conclusion The contents of diesters alkaloid in Radix Aconiti Kusnezoffii processed by three Mongolian distinct technology decrease largely,which can help to guide traditional Mongolian clinical medication.
