摘 要:马铃薯试管苗加入矮壮素(CCC)600mg/L于4℃低温保存150d,恢复生长后接种于不加任何激素的MS培养基上,在24℃的组培室进行恢复培养30d,长到3cm左右时,利用RAPD(Random amplified polymorphic DNA,随机扩增的多态性DNA)标记技术分析了其遗传稳定性。通过对120条随机引物进行筛选检测,发现马铃薯试管苗低温保存的过程中没有发生遗传物质的改变,在遗传性状上表现相对稳定,无明显的变异条带。Potato plantlets have been treated with MS + CCC of 600 mg/L and stored at 4℃ for 150 days at first,then they were inoculated to MS medium without any hormone,and then they renewal cultivated in tissue culture chamber at 24℃ for 30 days.When they approximately grew to 3 cm,their genetic stability was analyzed by RAPD(Random amplified polymorphic DNA) technology.Through detecting 120 random primers,the results showed that there had no genetic mutations during cryopreservation,the potato genetic character were relatively stable,and there are no apparent differential bands.
