摘  要:[目的]探索蕨麻猪生长激素基因与其生理学和生物学特性的关系。[方法]从蕨麻猪血液细胞中提取基因组DNA,通过PCR技术扩增出蕨麻猪生长激素基因序列,进行碱基组成多态性和突变分析。[结果]蕨麻猪生长激素(pGH)基因序列全长1671bp,有5个外显子和4个内含子。外显子1、2、3、4和5的碱基数依次是10、161、117、162和201bp,内含子1、2、3和4的碱基数分别是244、210、192和277bp。除第1外显子和第1内含子碱基剪切有明显的例外,其余全部符合GT-AG规则;蕨麻猪生长激素基因4种碱基含量依次为G>C>T>A。(G+C)含量和(A+T)含量分别在62%和37%以上。蕨麻猪编码区第4外显子在+1046位处A→G,为错义突变,其余均为同义突变。蕨麻猪生长激素基因编码的前体蛋白含216个氨基酸,成熟蛋白是由190个氨基酸残基组成的一个分泌性蛋白;二级结构中含有6个α螺旋结构,三级结构是结构松散的球状蛋白,含有6个α螺旋前区。[结论]为蕨麻猪生长激素基因序列的研究提供了参考依据和借鉴。[Objective] To explore the relation between growth hormone gene sequence with physiology and biology characteristic of Juema swine.[Method] The genome DNA was extracted from the blood of Juema swine.The growth hormone gene sequence of Juema swine was amplifyied through PCR and analyzed.[Result] The length of growth hormone gene sequence in Juema swine was 1 671 bp,and the gene sequence contained 5 exons and 4 introns.The base number of exons 1,2,3,4 and 5 were 10,161,117,162 and 201 bp,respectively,and the base number of introns 1,2,3 and 4 was 244,210,192 and 277 bp,respectively.GT-AG regulation can be found all the gene sequence except the first exon and intron.The order of four kinds bases content in Juema swine's growth hormone gene are G〉C〉T〉A.The contents of(G + C)and(A + T)are more than 62% and 37%,respectively.The forth exon of Juema swine showed missense mutation A→G in +1 046,others were all synonymous mutation.The precursor protein coding by Juema pig's growth hormone gene contained 216 amino acids,and maturation protein was a secretory protein consisting of 190 amino acid residue.It contained 6 α-helix helical structures in the secondary structure.It was a globular protein with loose tertiary structure,which contained 6 α-helix anterior regions.[Conclusion] The research provide reference and basis for researching the growth hormone gene sequence of Juema swine.
