摘  要:目的:探讨益母草水苏碱干预NE诱导乳鼠心肌细胞肥大的作用。方法:采用改良Simpson法培养乳鼠心肌细胞,选用NE诱导制作心肌细胞肥大模型,随机分为对照组,NE(10-6M)组,Sta高中低剂量(10-4M、10-5M、10-6M)组及Car(10-6M)组。在药物作用的1d、3d、5d和7d后收集细胞,采用图像分析系统测量心肌细胞表面积,BCA法检测细胞蛋白含量,荧光微量测定细胞DNA含量,ELISA法检测细胞ANP和BNP含量,以细胞表面积、Protein/DNA比值以及ANP和BNP含量作为检测指标,观察益母草水苏碱对肥大心肌细胞的药效学作用。结果:益母草水苏碱作用于细胞1d后,Sta大中小剂量组与NE组比较各项指标没有统计学差异。益母草水苏碱作用于细胞3d后,Sta大中剂量组与NE组比较可以明显降低细胞的表面积、Protein/DNA的比值及ANP含量(P<0.05),Sta小剂量组较NE组降低了细胞表面积(P<0.05),其它各项指标没有差异。益母草水苏碱作用于细胞5d后,Sta大中小剂量组较NE组显著降低细胞的表面积、Protein/DNA的比值及ANP含量(P<0.05),各组BNP含量与NE组比较,有降低的趋势,但是没有统计学差异。益母草水苏碱作用于细胞7d后,Sta大中小剂量组与NE组比较,明显降低细胞的表面积、Protein/DNA的比值、ANP和BNP含量(P<0.05)。结论:益母草水苏碱具有抗NE诱导乳鼠心肌细胞肥大作用。Objective:To observe the inhibitive effects of Stachydrine (Sta) on myocardial cell hypertrophy of neonatal rats induced by norepinephrine.Methods:Myocardial cells of neonatal rats were incubated with NE to establish myocardial cell hypertrophy models.Cultured myocardial cells of neonatal rat were randomized into the control group,NE (10-6M) group,Sta(10-4M,10-5M,10-6M)group,and Car(10-6M)group. After 1d,3d,5d and 7d of medical treatment,the cells were collected. The cell surface area of the myocardial cells were measured by image analytical system,the cell protein content were measured by BCA,and the cell DNA content were measured by fluorescent microdetermination,The contents of ANP and BNP were measured by ELISA. With cell surface area ,the ratio of Protein/DNA,and contents of ANP and BNP as the indexes of myocardial cell hypertrophy,the anti -hypertrophy effects of Sta were assayed. Results:There were no statistical differences in each index between the Sta groups with different concentrations and the NE group on the 1d. On the 3d,the cell surface area,ratio of Protein/DNA and content of ANP were significantly decreased in the 10-4M Sta group and 10-5M Sta group compared with those in the NE group(P0.05),while there were no statistical differences in BNP content between all groups. The cell surface area was decreased in the 10-6M Sta group compared with that in the NE group(P0.05),while there were no differences in other indexes. On the 5d,the cell surface area,ratio of Protein/DNA and content of ANP were significantly decreased in all the Sta groups compared with those in the NE group(P0.05). Although there were no statistical differences in BNP content between all groups,there was a tendency of decrease in BNP content. On the 7d,the cell surface area,ratio of Protein/DNA and contents of ANP and BNP were significantly decreased in all the Sta groups compared with those in the NE group(P0.05).
