摘 要:目的:研究高良姜不同提取物对小鼠学习记忆能力及胆碱能神经系统功能的影响,探讨其作用机制。方法:采用小鼠跳台法、小鼠Morris水迷宫法考察高良姜不同提取物对东莨菪碱所致记忆获得障碍小鼠的学习记忆能力的影响;紫外分光光度计检测小鼠大脑组织中乙酰胆碱转移酶(ChAT)及乙酰胆碱酯酶(TChE)的活力;HE染色观察海马组织形态学改变。结果:高良姜水及乙醇提取物组小鼠跳台潜伏期均显著延长(P<0.05),错误次数均显著减少(P<0.01),Morris水迷宫实验游泳潜伏期均显著缩短(P<0.05),TchE活力均显著降低(P<0.05),ChAT活力均显著增强(P<0.05),显著改善海马神经元的变性、脱落。结论:高良姜水及乙醇提取物均对东莨菪碱所致小鼠记忆获得障碍有显著改善作用,且高良姜水提取物的改善作用强,其作用机制可能与降低TchE活力、提高ChAT活力,增强中枢胆碱能神经系统功能有关。Objective:To explore the effects of different extract of Alpinia officinarum on learning and memory deficits and the function of the central cholinergic system in mice and compare with each other. Methods:Step-down test and morris water mase were performed to observe the learning and memory ability of mice impaired by scopolamine; The activity of ChAT and TChE were measured with colorimetry,The alterations in hippocampus slices morphology were assessed using microscope. Results:As compared with control group,the numbers of error was markedly increased (P0.01)and the step-down latency was markedly shortened (P0.05)in step-down test and the escape latency was markedly prolonged(P0.05) in Morris water mase,The activity of ChAT was markedly decreased (P0.05)and TChE was markedly increased (P0.05)in mice of model group; compared with model group,improved neuro degenerative changes and reduced the death of neuron in hippocampus slices,both water and ethanol extract of Alpinia officinarum could improve learning and memory deficits and the function of the central cholinergic system in mice impaired by scopolamine. Conclusion:These results indicated that both water and ethanol extract of Alpinia officinarum could markedly improve learning and memory deficits and the water extract of Alpinia officinarum is better than ethanol extract of Alpinia officinarum and the mechanism may be related with increasing the activity of ChAT and decreasing the activity of TChE and the improvement of the function of the central cholinergic system.
