中药PT液对衰老模型小鼠脑部凋亡细胞、Bcl-2和SOD的影响

摘　　要：目的:研究中药PT液对衰老模型小鼠脑部凋亡细胞、Bcl-2和SOD的影响。方法:将昆明种雄性小白鼠40只随机分成4组:正常对照组、模型组、小剂量治疗组(PT1)和大剂量治疗组(PT2)。8周后,断头处死,冰上取鼠的脑部,制成石蜡切片,做凋亡细胞的表达、Bcl-2的免疫组化染色和全血SOD的检测,随后计算凋亡细胞指数(AI)、Bcl-2阳性细胞率和做Bcl-2的免疫图像分析。结果:模型组的AI、Bcl-2阳性细胞率和SOD与对照组比较有显著性差异(P<0.05);PT1组的AI、Bcl-2阳性细胞率和SOD与对照组比较有显著性差异(P<0.05),PT2组的AI、Bcl-2阳性细胞率和SOD与对照组比较都无显著性差异(P>0.05);PT1组的AI、Bcl-2阳性细胞率和SOD与模型组比较无显著性差异(P>0.05);PT2组的AI、Bcl-2阳性细胞率和SOD与模型组比较有显著性差异(P<0.05)。结论:PT液对治疗衰老具有较好疗效,可通过抗氧化和调节Bcl-2来治疗凋亡,为临床中草药治疗衰老疾病中的凋亡提供理论依据。Objective：To study Chinese medicine PT liquid how to affect the apoptosis cell 、Bcl-2 and SOD in senescing little rats＇ brain.Methods：40 Male little white rats of Kunming species are randomized into 4 groups,normal control group（control group）,model group,little dose treating group（PT1） and big dose treating group（PT2）;after 8 weeks,executed by decollation.Then take out rats＇ brain on the ice,crank out paraffin slice and do the expression of apoptosis cell,immunity histochemistry dyeing of Bcl-2 and the measure of SOD in the blood,followed by calculating apoptosis cell index（AI）,calculating Bcl-2 masculine cell rate and doing immunity image analysis of Bcl-2.Results：AI,Bcl-2 masculine cell rate and SOD of model group is significant discrepant with control group（P〈0.05）;AI,Bcl-2 masculine cell rate and SOD of PT1 group is significant discrepant with control group（P〈0.05）,AI,Bcl-2 masculine cell rate and SOD of PT2 group are not significant discrepant with control group（P〉0.05）;AI,cl-2 masculine cell rate and SOD of PT1 group are not significant discrepant with model group（P〉0.05）;AI,cl-2 masculine cell rate and SOD of PT2 group are significant discrepant with model group（P〈0.05）.Conclusions：To treat senescing,PT liquid have good effect,can cure apoptosis by anti-oxidation and adjusting Bcl-2,which put forward the theory foundations on clinically treating apoptosis in senescing disease by Chinese herbal.medicine.
