摘 要:目的观察单用日本血吸虫(SJ)童虫细胞(SC)和联用免疫调节剂诱生小鼠免疫保护性的效果。方法制备感染后第18dSC;实验昆明小鼠分为8组:A组(PBS)、B组(SC)、C组(IL-2)、D组(IL-2+SC)、E组(IFN-γ)、F组(IFN-γ+SC)、G组(香菇多糖)、H组(香菇多糖+SC)。腹股沟皮下间隔2w免疫1次,共3次。于末次免疫第4w攻击感染,第35d后剖杀小鼠计数虫荷及每克肝卵数(LEPG),计算减虫率和LEPG减少率。ELISA测定第2、3次免疫后及攻击前小鼠血清中特异性抗童虫可溶性抗原(SWSA)的抗体。结果保护性效果显示:与A组比较,B、C、D、E、F、G和H组的减虫率分别为43.3%、13.5%、36.1%、32.7%、46.2%、18.3%和33.7%,LEPG减少率分别为59.8%、26.6%、52.1%、47.7%、57.7%、34.0%和53.7%。各免疫调节剂与SC联合免疫组与单用SC组比较,特异性抗体水平差异无统计学意义。结论SC能诱生小鼠较明显的免疫保护性,单用IFN-γ也有一定的减虫和减卵效果,但免疫增强剂均未能显著提高SC诱生的免疫保护性效果和体液免疫应答水平。Objective To observe protective immunity induced by Schistosoma japonicum (SJ) schistosomulum cells (SC) alone or Sj cells combined with different immunomodulators. Methods 18-day schistosomulum cells were obtained from rabbits infected with Sj cercaria. Kunming mice were divided into 8 groups:group A(PBS),B(schistosomulum cell,SC),C(IL-2),D (IL-2+SC),E(IFN-γ),F(IFN-γ+SC),G(Lentinan) and H(Lentinan+SC).Mice were immunized subcutaneously in inguinal groove for three times with 2 weeks interval. Four weeks after final immunization,mice were challenged with 30±1 cercarea. On the 35th day after challenge infection,mice were sacrificed and perfused,and the worm burden and liver egg load were counted. Specific antibodies against schistosomulum worm soluble antigens (SWSA) in sera from mice one day after second,third immunization and before challenge were detected by ELISAs. Results Compared with control group A,the reduction rate of worm burden were 43.3%,13.5%,36.1%,32.7%,46.2%,18.3% and 33.7% for group B,C,D,E,F,G and H,respectively;The reduction of liver eggs per gram were 59.8%,26.6%,52.1%,47.7%,57.7%,34.0% and 53.7% for group B,C,D,E,F,G and H,respectively.No significant differences in specific antibodies against SWSA between each group combined with different immunomodulators and SC group were found. Conclusion Significant immunoprotection induced by immunization with schistosomulum cells is observed. Immunization with IFN -γ also reveals certain protective immunity. Immunization with SC combined with immunomodulators does not significantly enhance the levels of immunoprotection and humoral immunoresponse than immunization with SC only.
