摘 要:以利用Trizol试剂盒提取的蝴蝶兰组培苗叶片总RNA为模板,根据已报道的建兰花叶病毒(Cymbidium mosaic virus,CyMV)和齿兰环斑病毒(Odontoglussm ringspot virus,ORSV)外壳蛋白基因(cp)的保守序列设计特异性引物,探索RT-PCR反应条件,建立了可同时检测CyMV和ORSV的双重RT-PCR体系。According to the reported Cymbidium mosaic virus (CyMV) and Odontoglussm ringspot virus (ORSV) coat protein (CP) gene sequences, two pairs of specific PCR primers for cp genes were designed respectively. And a dual RT-PCR method was successfully developed for detecting CyMV and ORSV at the same time with the template of total RNA isolated from the leaves of tissue culture plantlets by Trizol reagent kit. The dual RT-PCR method was confirmed as reliable and repeatable by applying to detect the two orchid viruses of several orchid species.
