PTEN在宫颈癌及宫颈内瘤样变组织中的表达及临床意义

摘　　要：目的探讨抑癌基因PTEN在宫颈鳞状上皮细胞癌和宫颈内瘤样变(CIN)组织中的阳性表达及相关性。方法用免疫组织化学(IHC)S-P法检测92例宫颈鳞癌及癌旁正常宫颈组织和40例CIN组织中PTEN的蛋白表达情况。结果宫颈鳞癌组PTEN阳性表达率为19.57%,CIN组为42.5%,癌旁正常宫颈组为97.83%,3组间比较差异均有统计学意义(P<0.05,P<0.01)。CINⅠ级、Ⅱ级、Ⅲ级PTEN的阳性表达率分别为66.67%、35.71%和18.18%,CINⅠ级与CINⅡ、Ⅲ级之间比较差异有统计学意义(P<0.05)。CINⅡ级、Ⅲ级间比较,差异无统计学意义(P>0.05)。PTEN在宫颈鳞癌组中的表达随TNM分期的上升而下降(P<0.05),而在不同年龄和组织学分级中差异无统计学意义(P>0.05)。结论随着宫颈病变程度的进展PTEN的表达呈下降趋势,提示PTEN表达降低是宫颈癌进展过程中的早期事件,也是宫颈组织恶变的信号。Objectives To explore the positive expression of PTEN protein in the tissues of cervical squamous cell cancer（CSCC） and cervical intraepithelial neoplasia（CIN）.Methods The protein expression of PTEN was detected by SP immunohistochemical stain（IHC） method in 92 cases of CSCC,normal tissues surrounding tissue, and 40 cases of CIN.Results The positive protein expression of PTEN in CSCC,CIN and normal tissue was 19.57%,42.5%and 97.83%,respectively.There vyere statistical difference among three groups（P0.01 or P0.05）.The positive protein expression of CINⅠ,CINⅡand CINⅢwas 66.67%,35.71%and 18.18%respectively. There was statistical between CINⅠand CINⅡ,CINⅢ（P0.05） groups,while no difference between CINⅡand CINⅢ（P0.05）.The expression of PTEN in CSCC was decreased with the progression of TNM stage,while no statistical difference in grading of age and tissue（P0.05）.Conclusion The protein expression of PTEN was gradually decreased with the development of CSCC,suggested that the decrease expression of PTEN was the indication of earlier stage of CSCC,as well as the signal of canceration of cervical tissue.
