摘 要:采用毛细管电泳-安培检测法(CE-AD),同时测定中药黄独中的表儿茶素、异香草酸、香草酸和杨梅素等四种主要生物活性成分的含量.实验采用柱端射壁式检测法,以直径300 m的碳圆盘电极为工作电极,检测电位为+0.95 V(vs.SCE).在优化的测定条件下,上述四种组分在40×10-3mol/L,pH8.7的硼酸盐缓冲溶液中15 min内达到基线分离.各组分在三个数量级的范围内呈良好线性关系,该方法成功的被应用于实际样品的分析,获得令人满意的结果.A method based on capillary electrophoresis with Amperometric detection has been developed for the separation and determination of epicatechin,isovanillic acid,vanillic acid,and myricetin in herb Dioscorea Bulbifera L for the first time.The effects of working electrode potential,pH and concentration of running buffer,separation voltage and injection time were investigated.Under the optimum conditions,the analytes could be separated in a 40 mmol borate buffer(pH8.7) in 15 minutes.A 300 m diameter carbon disk electrode has a good response at +0.95V(versus SCE) for all analytes.The response was linear over three orders.The method has been successfully applied to the analysis of real sample,with satisfactory results.
