摘  要:目的观察感染隐孢子虫大鼠CD4+/CD8+值的变化,为进一步探讨其免疫机制提供依据。方法将30只SD大鼠随机分为营养不良组、免疫抑制组和空白对照组。营养不良组采用8%低蛋白饲料喂养,免疫抑制组连续注射地塞米松,分别处理11d。从湖水中过滤得隐孢子虫卵囊,分2次灌胃营养不良组和免疫抑制组大鼠,每次灌胃量为3×103个/只,大鼠第2次感染隐孢子虫后1周处死。取小肠内皮组织,做病理切片,观察隐孢子虫感染情况;取脾脏、小肠黏膜下集合淋巴结及小肠内皮组织分别做成细胞悬液,用流式细胞检测仪检测T细胞亚类,计算CD4+/CD8+值。结果感染隐孢子虫免疫抑制组大鼠体重下降,脾脏严重发育不全;脾脏、小肠内皮及黏膜T淋巴组织中的CD4+/CD8+比值与营养不良组和空白对照组比较差异均有统计学意义(P<0.05)。结论感染隐孢子虫后,免疫抑制和营养不良大鼠较空白对照组大鼠CD4+/CD8+值降低更为显著。Objective To create a model of Cryptosporidiurn parvurn infection in immunosuppressed rats, detect helper T lymphocytes, and ascertain the effects of malnutrition and immunosuppression in a rat model. Methods Thirty Sprague-Dawley rats were randomly divided into 3 groups: a malnourished group, an immunosuppressed group, and a blank control group. The malnourished group was fed 8% low protein feed and the immunosuppressed group was given dexamethasone (DXM) for 11 days. Then, rats in the malnourished group and immunosuppressed group were orally inoculated twice with 3×10^3 oocysts of C. parvum that were acquired by filtering fresh water. All rats were sacrificed on the 7th day after the second infection. Intestinal endothelium was biopsied. Pathology specimens of oocysts were prepared to as certain the state of C. parvurn infection. Cell suspensions were prepared from the spleen, aggregated lymphatic follicles from the submucous layer of the intestine, and intestinal endothellum. The CD4^+/CD8^+ counts in cell suspensions were analyzed with flow cytometry. Results C. parvum infection led to a decrease in body weight in immunosuppressed rats and serious underdevelopment of the spleen. The CD4^+/CD8^+ ratio in the spleen, intestine, and mucosal T lymphoid tissue differed significantly (P〈0. 05) in comparison to that in the malnourished group and blank group. Conclusion In comparison to the blank controls, the CD4^+/CD8^+ counts in the other two groups decreased markedly after infection with oocysts of C. parvum.
