王老吉凉茶对拘束应激负荷小鼠脾脏CTL活性的影响

摘　　要：目的研究王老吉凉茶(Wanglaoji Herbal Tea,WHT)对拘束应激诱发小鼠脾脏细胞毒性T细胞(CTL)活性的影响。方法将雄性C57BL/6小鼠随机分为正常对照组、致敏对照、应激对照组、125及500mg/kg王老吉凉茶给药组,制备小鼠脾脏淋巴细胞悬液,流式细胞术分析T淋巴细胞亚群和CTL对P815细胞杀伤活性,同时检测小鼠脾脏淋巴细胞IL-2和IL-3的表达量。结果拘束应激明显降低Th淋巴细胞亚群的比例,显著降低CTL对P815瘤细胞的杀伤活性,同时减少脾脏淋巴细胞IL-2和IL-3表达量减少。与拘束应激组相比,王老吉凉茶给药可以增加拘束负荷小鼠脾脏淋巴细胞数目,提高CTL细胞杀伤活性,并且改善IL-2和IL-3表达量。结论王老吉凉茶对拘束负荷诱发小鼠免疫机能低下有改善作用。Objective To study the effect of Wanglaoji Herbal Tea（WHT）on cytotoxic T lymphocyte（CTL）activity in the spleen of restraint stress mice sensitized.Methods The male C57BL/6 mice were randomly divided into normalcontrol group,sensitized control group,stress control group,125 mg/kg and 500 mg/kg WHT groups.The spleen lymphocyte suspension of each group was prepared.The parameters of spleen T cell subsets,CTL activity was detected by flow cytometry,and the secretion of IL-2 and IL-3 in the spleen was detected by cytokine assay.Results Restraint stress depressed the number of TH cells,inhibited the activity of CTL against P815,and reduced the secretion of IL-2 and IL-3.Compared with the stress control group,125 mg/kg and 500 mg/kg WHT increased the number of lymphocytes,recovered the cytotoxicity of CTL,and restored the secretion of IL-2 and IL-3 in mice spleen.Conclusion Wanglaoji Herbal Tea has immunologic protective effect on immune function in restraint stress mice.
