摘 要:目的观察固本防哮饮对哮喘缓解期小鼠的影响。方法通过腹腔注射和雾化吸入卵蛋白(OVA)制备哮喘缓解期小鼠模型。在停止激发后给药4周,末次给药后24h观察小鼠气道反应性、肺泡灌洗液(BALF)中炎性细胞总数和细胞分类、白细胞介素-4(IL-4)和γ-干扰素(IFN-γ)水平以及肺脏病理学变化。结果固本防哮饮可以降低模型小鼠的气道高反应性,降低BALF中炎性细胞总数及嗜酸性粒细胞和中性粒细胞比例,升高IFN-γ水平,减轻支气管周围炎性细胞浸润,减轻气道重塑。结论固本防哮饮降低气道高反应性、减轻支气管周围嗜酸性粒细胞浸润以及调节机体IFN-γ水平可能是其防止哮喘复发的作用机制。Objective To observe the effects of Guben Fangxiao Decoction(GFD)on asthma mice at remission stage. Methods BALB/c mice were sensitized with intraperitoneal injection of OVA on day 1 and day 11,and with atomization inhalation of OVA on day 11.Sensitized mice involved with six 2-day periods of atomization inhalation of OVA challenges,and then the mice were administered intragastrically with GFD or distilled water for 4 weeks.The airway hyperresponsiveness,inflammatory cell count and classification in bronchoalveolar lavage fluid(BALF),interleukin-4(IL-4)and interferonγ(IFN-γ)levels in BALF and lung histological changes were observed 24 hours after last administration.Results GFD decreased the airway hyperresponsiveness,reduced BALF inflammation cell count and the ratio of eosinophile granulocytes and neutrophil granulocytes,increased IFN-γlevel in BALF,relieved the infiltration of inflammatory cells in the airway,and reduced airway remodeling.Conclusion The mechanism of GFD on preventing asthma attack might be related with the decrease of airway hyperresponsiveness and eosinophile granulocyte infiltration,and with the increase of IFN-γlevels.
