摘 要:目的观察葶苈生脉方对充血性心力衰竭(CHF)大鼠血浆血管紧素Ⅱ(AngⅡ)含量、心肌组织中AngⅡ1型受体(AT1)和传导因子蛋白激酶C(PKC)表达的影响,探讨该复方治疗CHF的作用机制。方法采用腹主动脉缩窄法复制CHF大鼠模型,实验分假手术组,模型对照组,葶苈生脉方高、低剂量组,阳性药心宝丸对照组。测定血浆中AngⅡ含量,检测心肌组织AT1受体和传导因子PKC的表达。结果与假手术组比较,模型组大鼠血浆AngⅡ的含量明显升高(P<0.01),心肌组织中AT1、PKC表达显著增加(P<0.01);经药物干预后,各治疗组AngⅡ的含量均显著降低(P<0.01),心肌组织AT1、PKC的表达明显降低(P<0.01)。结论葶苈生脉方能降低心衰大鼠血浆中AngⅡ的含量,下调心肌组织中AT1、PKC的表达,以达到拮抗心力衰竭的作用。Objective To explore the therapeutic mechanism of Tingli Shengmai Decoction(THD)for congestive heart failure(CHF)by observing its effects on AngⅡand its signal conducts in CHF rats.Methods CHF animal models were established by constriction of the abdominal aorta,and rats were randomly devided into sham-operation group,model group,high-and low-dose THD groups,and positive control(Xinbao Wan)group.The AngⅡcontent in the plasma,the AT1 receptor expression and signal conduct factor PKC expression were detected.Results Compared with those of the sham-operation group,the plasma AngⅡcontent,and the expression of AT1 receptor and signal conduct factor PKC in myocardial tissue of the model group were significantly increased(P〈0.01).Compared with those of the model group,the plasma AngⅡcontent,and the expression of AT1 receptor and signal conduct factor PKC in myocardial tissue of THD groups were significantly decreased(P〈0.01).Conclusion Tingli Shengmai Decoction can decrease the AngⅡcontent in the plasma and down-regulate the expression of AT1 receptor and signal conduct factor PKC in myocardial tissue of CHF rats,which probably contributes to the therapeutic mechanism of Tingli Shengmai Decoction.
