摘 要:目的对浙江七叶树种子娑罗子中七叶皂苷的纯化工艺进行研究,以获得最佳纯化工艺和高纯度的七叶皂苷。方法采用超声波法提取七叶皂苷,以D101大孔吸附树脂分离纯化七叶皂苷,并以吸附率和洗脱率为指标优化纯化工艺,D101大孔吸附树脂最佳工艺再配合硅胶柱层析精制七叶皂苷。应用紫外分光光度法跟踪检测七叶皂苷含量,评价纯化效果。结果 D101大孔吸附树脂纯化七叶皂苷的最佳工艺为:静置吸附24h,上样液浓度为30mg/mL,上样速度为2BV/h(树脂床体积),洗脱剂为70%乙醇,洗脱体积为2.5BV,洗脱速度为1.5BV/h。D101大孔吸附树脂最佳工艺联合硅胶柱层析,七叶皂苷纯度可达90%以上。结论此纯化工艺稳定可行,可有效用于浙江七叶树种子娑罗子中七叶皂苷的分离纯化和制备。Objective To study the separation and purification technology of escins,which extracted from the seeds of A.chinensis Bge. var. chekiangensis(Hu et Fang)Fang(Semen Aesculi),thus to screen the optimal purification technology of escins.Methods Escins were extracted by ultrasonic extraction,the adsorption rate and desorption rate were taken as the indexes to optimize the purification technology of escins by using macroporous absorption resin D101. Macroporous absorption resin D101 combined with silica gel column chromatography was used to prepare escins.The separating efficiency was evaluated by measuring the concentration of total escins with ultra-violet spectrophotometer. Results The optimum purification condition for escins with macroporous absorption resin D101 was as follows:the concentration of extract sample reached 30 mg/mL,the current velocity of adsorption was 2 BV/h(resin bed volume),2.5 BV 70%ethanol was used as eluant and the eluted velocity was 1.5BV/h.The combination of macroporous absorption resin D101 with silica gel column chromatography increased the purity of escins to more than 90%.Conclusion The established method is simple and feasible,and can be used for the separation and preparation of escins from the seeds of A.chinensis Bge. var. chekiangensis(Hu et Fang)Fang(Semen Aesculi).
