摘 要:利用聚丙烯酰胺凝胶电泳研究了金属模拟酶铜(Ⅱ)配合物对肌红蛋白水解反应的影响;测定了不同空间位阻、缓冲溶液和pH值条件下的水解效率.结果表明:水解位点位于Gln91-Ser92和Ala94-Thr95,显示铜(Ⅱ)配合物对肌红蛋白的水解具有很高的选择性;与此同时,构型不同的铜(Ⅱ)配合物对肌红蛋白的水解影响程度存在差异.磷酸盐和碳酸氢铵可加强水解作用,氯离子使水解作用减弱,而在pH6.5~8.5范围内水解效率最高.The effect of Cu(Ⅱ) complex with artificial metallopeptides on hydrolyzation of myoglobin was studied by means of polyacrylamide gel electrophoretogram.The hydrolytic efficiency under different conditions of spacial barrier,buffer and pH values was measured.Results indicate that two hydrolytic sites at Gln91-Ser92 and Ala94-Thr95 are involved in the hydrolyzation process of myoglobin,which indicates that the Cu(Ⅱ) complexes have a high selectivity to hydrolyzation of myoglobin.At the same time,Cu(Ⅱ) complexes with different configurations have different extent of effect on the hydrolyzation of myoglobin.Phosphate and ammonium bicarbonate can enhance the hydrolyzation,but chloride retards the hydrolyzation;and the highest rate of hydrolysis is obtained at pH 6.5~8.5.
