HPLC-ELSD同时测定平贝母中贝母素甲和贝母素乙含量

摘　　要：目的建立同时测定中药材平贝母中贝母素甲、贝母素乙含量的测定方法。方法采用Agilent TC-C18(4.6mm×250mm,5μm)色谱柱,以甲醇-乙腈-0.5%醋酸溶液(8:19:73)为流动相,流速为0.6mL·min-1,蒸发光散射检测器检测。结果贝母素甲、乙的线性范围分别为0.400～3.200mg·mL-1(r=0.9998)和0.304～2.432mg·mL-1(r=0.9999),平均回收率分别为101.6%和102.6%。结论该方法便捷、有效,适用于检测贝母类生物碱等无紫外吸收的化学成分。Objective To establish a method for determination the contents of peimine and peiminine in Bulbus fritiliariae ussuriensis.Methods Agilent TC- C18 （4.6 mm×250 mm,5 μm） was used,with ELSD detected,the methanol-acetonitrile-0.5%acetic acid solution（8：19：73）as mobile phase,the flow rate was 0.6 mL·min-1.Results The linear ranges of peimine and peiminine were in 0.400～3.200 mg·mL-1 （r=0.999 8）and 0.304～2.432 mg·mL-1 （r=0.999 9） ,the average recoveries（n=5）were101.6% and 102.6%.Conclusion The method is convenient and effective,it can be used for determination of chemical constituents without ultraviolet absorption.
