摘 要:目的:探讨脂多糖(1ipopolysaccharide,LPS)对人近曲肾小管上皮细胞(HK-2)表面活性蛋白D(surfactant protein D,SP-D)及肿瘤坏死因子(TNF-α)表达的影响。方法:将体外培养HK-2细胞分为5组,分别给与0、0.1、1、5、10μg/mL LPS刺激8h。免疫印迹和RT-PCR观察SP-D蛋白及mRNA表达变化,ELISA和RT-PCR观察TNF-α蛋白和mRNA表达变化。结果:1、5、10μg/mL LPS刺激8h可引起HK-2细胞SP-D蛋白及mRNA表达显著降低(P<0.05),1、5、10μg/mL LPS刺激8h可引起HK-2细胞TNF-α蛋白及mRNA表达显著增加(P<0.05)。SP-D与TNF-α的表达呈负相关。结论:LPS刺激下TNF-α表达增加与SP-D的表达降低有关。SP-D可能在肾脏炎症反应中发挥重要作用。Objective To investigate the effects of 1ipopolysaccharide (LPS) on the expressions of surfactant protein D (SP-D) and TNF-α in human renal proximal tubular epithelial cells (HK-2). Methods HK-2 cells were treated with LPS at various concentrations(0,0.1,1,5 and 10 μg / mL) for 8 h. Expression of SP-D was detected by Western blot and RT-PCR and that of TNF-α was determined by ELISA and RT-PCR. Results The expression levels of SP-D mRNA and protein in HK-2 cells significantly decreased after treatment with LPS at 1,5,and 10 μg / mL (P 0.05) while those of TNF-α mRNA and protein increased remarkably (P 0.05). The expression of SP-D was negatively associated with that of TNF-α. Conclusions With LPS stimulation,an increase in the expression of TNF-α is associated with a decrease in the expression of SP-D. SP-D may play an important role in the modulation of renal inflammation.
