摘  要:目的:探讨积雪草苷对盲肠结扎穿孔(cecal ligation and puncture,CLP)所致小鼠脓毒症诱导的急性肾损伤的影响。方法:48只昆明小鼠随机分为假手术组、CLP模型组、CLP+积雪草苷15,45 mg.kg-1治疗组。记录一般情况及24 h死亡率,并于手术后24 h,试剂盒测血清尿素氮(urea nitrogen,BUN)、肌酐(creatinine,Cr),酶联免疫法(ELISA)测血清白介素6(interleukin-6,IL-6),免疫印迹法检测肾脏诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)蛋白含量,并取肾脏组织作组织学分析。结果:与CLP模型组相比,积雪草苷降低了脓毒性小鼠死亡率,且剂量依赖性地减轻肾脏组织损伤,降低了血清BUN,Cr,IL-6水平及肾脏中iNOS蛋白含量。结论:积雪草苷对脓毒症致急性肾损伤具有保护作用,该作用可能与其降低血清IL-6及肾脏中iNOS蛋白含量有关。Objective:The present study is to investigate the protective effects of asiaticoside on sepsis-induced acute kidney injury in mice.Method:With the sepsis induced by cecal ligation and puncture(CLP),forty eight kunming mice were randomly divided into four groups as sham operated,CLP treated,CLP+asiaticoside 15,45 mg·kg-1 groups.General conditions and the amount of dead rate of mice were observed.The BUN and Cr levels were observed by the kits.IL-6 in serum was assayed by enzyme-linked immunosorbent assay(ELISA).Kidney tissues were harvested for determination of iNOS expression by Western blotting analysis.The pathologic changes were observed under electron microscope via hematoxylin-eosin(HE)stain.Result: Compared with CLP group,the death rate,the levels of BUN,Cr,IL-6,and iNOS protein expression of asiaticoside groups were significantly reduced.The pathologic changes in kidney tissues induced by sepsis were significantly attenuated dose-dependently by asiaticoside under electron microscope.Conclusion:Asiaticoside has protective effects against sepsis-induced acute kidney injury,which were probably associated with the inhibition of IL-6 in serum and iNOS protein in kidney tissues.
