摘 要:以生命科学中提出的对分离和定量分析有较高要求的研究课题为目标,将迎头分析(FA)与毛细管区带电泳(CZE)相结合,运用毛细管区带电泳/迎头分析(CZE/FA)技术,进行碱性药物propranolol与人血清白蛋白(HSA)平衡体系中相互作用参数的研究.A capillary zone electrophoresis/Frontal Analysis method was discussed and applied to the study of association parameters of a basic drug, propranolol (PRO), to human serum alhumin (HSA) tinder drug-HSA interaction equilibra. After the direct hydrodynamic injection (110 kPa· s) of a drug-HSA mixture into a coated capillary (effective length, 32 cm; 50 μm i. d. ) , the basic drug is eluted as a zonal peak with a plateau region under condition of phosphate buffer ( pH 7.4, ionic strength = 0.17) at 10 kV running voltage. The unbound concentration of drug can be detennined from the plateau peak height with good linear response, r = 0.9991 ( n = 8) . Eight PRO-HSA solutions (containing 100 - 450 μmol/L of PRO and 400 - 50 μmol/L of HSA, respectively) were investigated. The obtained data were processed with Scatchard linear regression analysis and the final estimated PRO-HSA association parameters include binding sites n =0.58 ( ±0.03) and binding constant K = 1500( ± 110).
