摘 要:目的了解糖尿病模型大鼠心肌组织中心肌营养素-1(CT-1)蛋白的表达,探讨黄芪注射液改善糖尿病大鼠心肌病变的作用机制。方法雄性Wistar大鼠随机分为正常组8只及糖尿病造模组22只,糖尿病造模组成模后再随机分为模型组和黄芪组,每组11只。黄芪组予黄芪注射液8 g/(kg.d)灌胃8周。测定大鼠心脏指数和心肌纤维含量的变化,并用免疫组化法检测CT-1的表达。结果模型组和黄芪组血糖高于对照组(P<0.01);心脏指数(心脏重量/体重)模型组高于正常组(P<0.01),黄芪组心脏指数较模型组降低(P<0.05);Masson染色中以目标面积计算的胶原含量,模型组高于正常组(P<0.01),黄芪组比模型组减少(P<0.05);CT-1蛋白的表达模型组高于正常组(P<0.01),黄芪组较模型组减少(P<0.05)。结论 CT-1的过度表达参与了糖尿病心肌病变中心肌肥大的发病过程,黄芪注射液对CT-1的表达和心肌肥大有抑制作用。Objective To study the protein expression of myocardial cardiotrophin-1 (CT-1) and mechanism of Huangqi Injection for relieving myocardial pathological changes in diabetic rats. Methods All male Wistar rats were randomly divided into the normal group (n = 8) and diabetic model group (n = 22), and the later was randomly divided again into the model group and Huangqi group ( each n = 11 ) after modeling. The Huangqi group was given intragastrically Huangqi Injection (8 g/kg ~ d) for 8 weeks. The pathological changes of heart index (heart weight/body weight ) and myocardial fiber content were determined and the expression of CT-1 was detected by using immunohistochemistry. Results The level of plasma glucose was higher in the model group and Huangqi group than that in the normal group (P 〈 0.01 ). The heart index was higher in the model group than that in the normal group (P 〈0.01 ), and was lower in the Huangqi group than that in the model group (P 〈 0.05 ). The content of collagen calculated by target area in Masson staining was higher in the model group than that in the normal group (P 〈0.01 ), and lower in the Huangqi group than that in the model group (P 〈 0.05 ). The expression of CT-1 was higher in the model group than that in the normal group (P 〈 0.01 ), and lower in the Huangqi group than that in the model group ( P 〈 0.05 ). Conclusion The excessive expression of CT-1 takes part in the myocardial hypertrophy in diabetic myocardial disease. Huangqi Injection has the inhibitory effects on the expression of CT-1 and myocardial hypertrophy.
