摘  要:目的:探讨烫伤创面愈合过程中MMP-2和TIMP-1表达的相关性。方法:在8只大鼠背部制成20%TBSA深Ⅱ度烫伤模型,烫伤后分别在24小时,4,7,14,20,29,35,62天时切取每只大鼠背部创面标本,采用免疫组化方法检测标本中MMP-2及TIMP-1的表达情况;在实验中取2只正常大鼠皮肤标本,同样检测标本中MMP-2及TIMP-1的表达情况。结果:MMP-2和TIMP-1在正常大鼠皮肤中几乎无表达;在大鼠烫伤后不同时期表达强度不同,24小时后开始表达,以后逐渐增强,大约到第20天达到高峰,持续约15天后减弱;到创面愈合后仍有表达。MMP-2的表达水平与TIMP-1的表达水平呈正相关。结论:MMP-2和TIMP-1的表达水平在创面愈合过程中有一定规律,与烫伤创面的愈合和瘢痕的形成可能有密切关系。Objective:To investigate the correlation between expression of MMP-2 and TIMP-1 in the process of scald wound repairing.Methods:Backs of eight big rats were scalded into 20% TBSA deep partial thickness burn.Biopsies from wound skins of each rat harvested at 24PBHS and 4,7,14,20,29,35,62PBDS.The expressions of MMP-2 and TIMP-1 in the samples were detected immunohistochemically.MMP-2 and TIMP-1 were also detected in the skin of another 2 normal rats as normal contro1.Result:MMP-2 and TIMP-1 were hardly expressed in the normal skins of rats.After rats were scalded,MMP2 and TIMP-1 had different expressions in the process of scald wound repairing.MMP-2 and TIMP-1 began to express at 24 PBHS,and enhanced gradually,and peaked on 20PBDS,and continued for 15 days then reduced;but MMP-2 and TIMP-1 were still expressed up to wound healing.There were correlations between the expression of MMP-2 and the expression of TIMP-1.Conclusion:The expression levels of MMP-2 and TIMP-1 had certain laws during wound repairing,which were closely related to wound repairing and the process of scar tissue growing.
