摘 要:建立了血中的苯妥英钠和苯巴比妥钠的胶束电动毛细管电泳分析方法(MEKC)。以25mmol/L硼酸钠溶液(pH=8.60)、30mmol/L十二烷基硫酸钠(胶束相)和甲醇30%(φ)为缓冲液,恒定电压25kV(电流30~35μA),检测波长200nm,毛细管柱长37cm。苯妥英钠和苯巴比妥钠两种药物质量浓度的线性范围为1.0~80mg/L,最低检出质量浓度为1.0mg/L。该法为临床提供了一种高效。The determination of phenytoin sodium and phenobarbital sodium by micellar electrokinetic capillary chromatography(MEKC) with sodium dodecyl sulphate(SDS)as the micellar phase is described. The factors affecting the micellar electrokinetic separation were studied for quantitative determination of two components in standards and in blood. The optimal separation of two components was achieved with 25 mmol/L borate sudium buffer(pH 8.60 ),30 mmol/L SDS solution, 30% methanol as a buffer and a constant voltage of 25 kV(current of 30~35 μA). The method has a good linearity in the range of 1.0~80 mg/L of the drugs and detection limit of 1.0 mg/L. Thus, the electrokinetic capillary chromatography is demonstrated to be a new, rapid, highly effective method for analysing the drugs in human blood quantitatively.
