白花蛇舌草对肝损伤小鼠T细胞亚群、TNF-α和IL-6的影响

摘　　要：目的探讨白花蛇舌草治疗CCl4所致肝损伤小鼠的免疫机制。方法 40只健康小鼠被随机分为四组:对照组每天灌胃等量生理盐水;损伤组在实验的第1天和第5天腹腔注射CCl4,同时每天灌胃等量的生理盐水;治疗组在实验的第1天和第5天腹腔注射CCl4,同时每天灌胃白花蛇舌草水提取物。以上各组均连续实验7d,在实验第8天摘眼球取血,用流式细胞仪检测小鼠外周血T细胞亚群,用放免分析法检测血浆中TNF-α和IL-6的含量,并进行肝组织常规病理学检查。结果白花蛇舌草可显著减轻肝组织病理损伤程度,提高外周血CD4+T细胞的百分比和CD4+T细胞/CD8+T细胞的比值,降低CD8+T细胞的百分比,降低血浆中TNF-α和IL-6的水平。结论白花蛇舌草调整CD4+T细胞和CD8+T细胞之间的平衡,降低血浆TNF-α和IL-6的水平,这可能是其治疗肝损伤的机理之一。Objective To investigate the immune mechanism of Hedyotis Diffusa Willd（HDW）for treating mice liver injury led by CCl4.Methods 40 healthy adult mice were randomly divided into four groups：Control group was given same volume normal saline by I.g every day;Injury group was given CCl4 by i.p.injection on the first and the 5th day,and it was given same volume normal saline by I.g every day;Treatment groups were given CCl4 by i.p.injection on the first and the 5th day,and they were given HDW water soluble extracts by I.g every day.Those measures were taken for seven days.On the 8th day,after blood samples were collected in each group,the T-lymphocytes subgroup was detected by FCM and the levels of TNF-α,IL-6 in plasma were detected by radioimmunoassay.The pathological change of liver tissue was inspected.Results HDW could significantly relieve pathological damage in liver.Compared with model group,the percentage of CD4＋ T-lymphocytes and the value of CD4＋ T-lymphocytes /CD8＋ T-lymphocytes of mice in peripheral blood in HDW treatment groups increased,on the other hand the percentage of CD8＋ T-lymphocytes and the levels of TNF-α,IL-6 in HDW treatment groups decreased.Conclusion HDW can regulate the balance between CD4＋ T-lymphocytes,CD8＋ T-lymphocytes and decrease the levels of TNF-αand IL-6 in plasma.It is possibly one of mechanism for HDW treating liver injury.
