摘  要:目的探讨体外人巨细胞病毒(HCMV)感染对人涎腺导管上皮细胞(HSG)角蛋白表达的影响。方法应用免疫细胞化学法检测体外培养的HSG中角蛋白8和18(K8和K18)的表达;蛋白印迹法检测HCMV感染对体外培养的HSG中K8和K18表达的影响;半定量逆转录聚合酶链式反应法检测HCMV对体外培养的HSG中K8和K18转录水平的影响。结果体外培养的HSG中K8和K18呈阳性表达;HCMV感染体外培养的HSG 0~96 h,K8的表达呈逐渐下降趋势,K18的表达在感染后24h内表达升高,此后下降;K8和K18的mRNA水平和蛋白水平均呈反向平行关系。结论 HCMV感染体外培养的HSG可引起角蛋白表型发生改变,K8和K18表达水平改变的调节发生在转录后水平。Objective To investigate the effects of human cytomegalovirus (HCMV) infection on keratin expression of human salivary gland duct cell (HSG) in vitro. Methods Expressions of keratin 8 and 18 (K8 and K18) were detected in HSG cultures in vitro by immuocytochemistry method. Effects of HCMV infection on the expressions of K8 and K18 of HSG in vitro were investigated by Western blotting. Semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR) was used to evaluate the transcription levels of K8 and K18 of HSG infected by HCMV. Results K8 and K18 were positively expressed in HSG cultured in vitro. The expression of K8 showed a decreased tendency from 0 h to 96 h post-infection while that of K18 at 45 kD increased during 24 h post-infection and reduced during the rest of time. The mRNA levels were paralleled reversely with protein levels of K8 and K18 of HSG. Conclusion Keratin phenotypic alterations are induced in HSG in vitro by HCMV infection. Expression level alterations of K8 and K18 of HSG are regulated at post-transcriptional levels.
