摘  要:目的:探索气虚证在大鼠垂体基因表达层面的物质基础。方法:采用16周龄Wistar大鼠、自发性高血压大鼠(SHR大鼠)、自发性糖尿病大鼠(GK大鼠),综合应用标准化大鼠诊法,及GeneChip Rat Gene1.0ST Array等技术,检测正常Wistar大鼠、正常气虚Wistar大鼠、SHR气盛大鼠、SHR气虚大鼠、GK气盛大鼠、GK气虚大鼠等垂体基因的表达,重点关注3个气虚证垂体基因表达较正常对照一致下调的基因。结果:共筛选得到24个基因,(1)与基因转录、翻译和修饰有关的基因8个:Leo1、Aatf、Myst2、Ttc12、Mtfmt、Sdccag10、Nmt2、LOC364236;(2)与信号转导、细胞活动、细胞生命有关的基因10个:Plce1、Sgk1、Sema3a、Nrp1、Tagln、Rnf185、Arl5b、Pcdhb20、Stk17b、Sept1;(3)不属于以上两类的6个:Fdft1、Abca5、Aox1、Ccdc93、RGD131055、AB086234(mRNA号)。结论:气虚大鼠垂体有其特征性的下调基因群,其具体的调控机制及生理/病理意义,以及与那些上调基因群的关系,有待深入的研究。Objective:To explore the material base of qi deficiency in pituitary gene expression of rats.Method:Sixteen weeks old Wistar rats,Spontaneously hypertensive rats(SHR rats) and Spontaneous diabetic rats(Goto-Kakizaki rats,GK rats) were used. By the Standardized quantitative four diagnosis methods and GeneChip Rat Gene 1.0 ST Array,pituitary gene expression of normal Wistar rats and normal Wistar rats with qi deficiency,SHR rats with qi deficiency,GK rats with qi deficiency were measured.Focus on the genes whose express down-regulated consistently in three qi deficiency syndromes compared to normal Wistar rats.Results:Twenty-four genes were gained,(1) Eight genes are related with transcript,translation and modification of genes:Leo1 Aatf Myst2 Ttc12 Mtfmt Sdccag10 Nmt2 LOC364236; (2) Ten genes are related with signal transduction,cell activity and cell life:Plce1 Sgk1 Sema3a Nrp1 Tagln Rnf185 Arl5b Pcdhb20 Stk17b Sept1; (3) Six genes don't belong to the functions above:Fdft1 Abca5 Aox1 Ccdc93 RGD131055 AB086234(mRNA ID). Conclusion:There are distinctively down-regulated genes in pituitary of qi deficiency rats,but their regulatory mechanism,the physiological and pathological signification and the relationship with up-regulated genes are awaited to deep research.
