摘 要:目的探讨产生内源性一氧化碳(carbon monoxide,CO)的关键酶血红素氧合酶-2(heme oxygenase-2,HO-2)在新生大鼠延髓的表达及意义。方法采用新生Sprague-Dawley大鼠,行石蜡切片,分别进行HE染色和免疫组织化学染色(SP法),观察新生大鼠延髓神经核团的分布和不同神经核团中神经元的形态,以及新生大鼠延髓是否存在HO-2阳性神经元和阳性神经元的分布情况。结果在舌下神经核、疑核、巨细胞网状核、面神经核等部位均可见HO-2阳性神经元。结论 HO-2阳性神经元存在于新生大鼠延髓呼吸相关神经核团,提示内源性CO可能参与中枢性呼吸活动的调节。Objective To test the expression and significance of heme oxygenase-2(HO-2),a key enzyme of synthesizing carbon monoxide(CO),in the medulla oblongata of newborn rats.Methods The experiments were carried out on the medullary slices of newborn Sprague-Dawley rats with paraffin section,the distribution of nuclei and the morphology of neurons in the medulla oblongata of newborn rats were observed with HE staining,the expression of HO-2 in the medulla oblongata of newborn rats was investigated with the immunohistochemical staining.Results HO-2-positive immunoreactive neurons could be observed in hypoglossal nucleus,ambiguous nucleus,gigantocellular reticular nucleus and facial nucleus in the medulla oblongata of newborn rats.Conclusion HO-2-positive immunoreactive neurons exist in the medullary respiratory related nuclei of newborn rats,which suggests that endogenous CO may be involved in the regulation of central respiratory activity.
