鸡传染性支气管炎病毒ZZ2004分离株3a、3b及E基因的序列测定与分析

摘　　要：采用随机引物PCR技术从具有腹泻、呼吸道症状及肾脏病变的肉鸡及无明显症状但生长迟缓的鸭体内获得一段长1171bp的片段,将此片段克隆入pMD18-T载体,测序后将序列用网上的BLAST软件和DNASTAR软件进行分析,结果表明所测序列与IBV主要参考毒株的同源率为82.0%-94.4%,与国内主要毒株CK/CH/LGD/04III、CK/CH/LGD/04II、chicken/JS/YZ07/2008、CK/CH/LSC/99I、SAIBK的同源率较高,达92.4%-94.4%,与国外毒株M41、H120、Beaudette、IBV4/91的同源性较低达85.2%-87.9%,从而确定所分离毒株ZZ2004为肾型传染性支气管炎病毒。The PCR product of length of 1 171 bp was derived from chickens which had,respiratory symptom and pathological changes of the kidneys and ducks which had no obvious symptom by using random PCR,and was cloned into pMD18-T vector.and then was analyzed using the software BLAST and DNASTAR.The result showed that the identity of the nucleotide sequence of obtained gene between ZZ2004 isolate and other IBV reference strains were from 82.0%to 96.0%,and the identity of that and other domestic IBV reference strains were from 92.4%to 94.4%.The identity of that and other overseas IBV reference strains were from 85.2%to 87.9%.The present study indicated that ZZ2004 isolate may be a member of avian nephrotic infectious bronchitis virus.
