摘 要:目的探讨酸味中药复方防治糖尿病血管病变的机制。方法采取腹腔注射小剂量链脲佐菌素配合高热量饮食的方法复制2型糖尿病大鼠模型。将大鼠随机分为正常组、模型组、氨基胍(AG)组和中药组。给药8周和12周末处死大鼠,采用荧光法和原位杂交法分别检测大鼠主动脉晚期糖基化终产物(advancedglycation end products,AGEs)含量和细胞间黏附因子-1(intercellular adhesion molecule-1,ICAM-1)基因表达。结果模型组大鼠主动脉AGEs含量和ICAM-1基因表达量均显著高于正常组(P<0.05);中药组和AG组主动脉AGEs含量和ICAM-1基因表达量显著低于模型组(P<0.05)。结论酸味中药复方可能通过降低糖尿病大鼠主动脉AGEs含量、下调ICAM-1 mRNA的过度表达达到延缓和治疗糖尿病血管病变的目的。Objective To investigate the possible mechanism of Compounded Sour Medicinal Herbs (CSMH) in preventing and treating type 2 diabetic mellitus (T2DM) induced macroangiopathy. Methods The rat model of T2DM was induced by using intraperitoneal injection of low dosage streptozotocin (STZ) and high-fat and high-caloric diets. The rats were randomly divided into normal control group, model group, aminoguanidine (AG) treated group and CSMH treated group. Eight and twelve weeks after the administration, the rats were sacrificed for assaying the level of advanced glycation end products (AGEs) and the expression of intercellular adhesion molecule 1 (ICAM-1) mRNA in the rat aorta tissues using fluorometrie method and in situ hybridization, respectively. Results The level of AGEs and the expression of ICAM-1 mRNA in the aorta tissues of rats in model group were significantly increased compared with those in normal group; and both of them were significantly decreased in CSMH and AG treated groups compared with those in model group. Conclusion CSMH might delay and treat T2DM in- duced macroangiopathy by decreasing the level of AGEs in the aorta tissues and down-regulating the over expression of ICAM-1 mRNA.
