补肾健脾颗粒对骨性关节炎家兔关节软骨蛋白聚糖含量及病理变化的影响

摘　　要：目的观察补肾健脾颗粒对实验性骨性关节炎家兔关节软骨蛋白聚糖含量及软骨病理组织学的影响。方法采用膝关节腔内注射木瓜凝乳蛋白酶法复制家兔骨性关节炎模型,灌胃给予小(2.0 g/kg)、中(4.0 g/kg)、大(8.0 g/kg)剂量补肾健脾颗粒进行治疗及其中剂量(4.0 g/kg)进行预防,模型复制14 d末取关节承重部位指定区域的关节软骨进行蛋白聚糖含量的测定和病理组织学检查。结果与模型组比较,补肾健脾颗粒预防组,补肾健脾颗粒中、大剂量组关节软骨蛋白聚糖含量显著增加;补肾健脾颗粒大剂量组和预防组关节软骨病理组织学变化明显改善。结论补肾健脾颗粒可对抗因木瓜凝乳蛋白酶诱导的骨性关节炎模型家兔软骨中蛋白聚糖的缺失,对其病理组织学改变有一定的治疗和预防作用。Objective To observe effects of Bushen Jianpi Granules, a traditional Chinese pr invigorating the kidney and the spleen, on histopathologic changes of cartilage and contents of escription for proteoglycan in articular cartilage of experimental rabbits with osteoarthritis （OA）. Methods OA model in rabbits was induced by injecting chymopapain into the articular cavity of the knee. The model rabbits were treated by gavage with low （2.0 g/kg）, middle （4.0 g/kg）, and high （8.0 g/kg） dosage Bushen Jianpi Granules; meanwhile, another group of rabbits were administered with middle-dosage （4.0 g/kg） for prophylaxis. The contents of proteoglycan in articular cartilage on the designated weight bearing area were determined and the histopathology of cartilage was examined 14 days after model being induced. Results Compared those in the model rabbits, the contents creased in the rabbits treated with middle- and of pro high-d teoglycan in articular cartilage were significantly in- osage Bushen Jianpi Granules as well as in the rabbits administered with middle-dosage for prophylaxis. The histopathologic changes of cartilage were also signif- icantly ameliorated in groups treated with high-dosage Bushen Jianpi Granules and middle-dosage for prophylaxis. Conclusion Bushen Jianpi Granules could antagonize the deficiency and loss of proteoglycan in articular cartilage of OA rabbits induced by chymopapain, and prevent histopathologic changes of cartilage.
