摘  要:目的研究乌头碱对体外培养的卵巢黄体细胞的毒性。方法无菌下制备25~29日龄雌性SD大鼠的卵巢黄体细胞,并通过透射电镜观察管嵴状线粒体来鉴定细胞。用MTT法检测0、0.05、0.5、5、50μg·mL-1的乌头碱对大鼠卵巢黄体细胞活力、激素分泌功能的影响,及对大鼠黄体细胞的氧化损伤作用。结果与溶剂对照组比较,作用24h后,50μg·mL-1乌头碱可明显抑制大鼠黄体细胞的增殖,差异有统计学意义(P<0.01);0.5、5、50μg·mL-1乌头碱对孕酮的分泌明显低于对照组(P<0.05)。结论乌头碱浓度高于5×102ng·mL-1时,对雌性大鼠卵巢黄体细胞有毒性作用,主要表现为抑制黄体细胞的增殖及抑制激素的分泌。OBJECTIVE To study the toxic effects of aconitine on rat lutein cells in vitro.METHODS Lutein cells were isolated from ovaries of 25-29 day-old female SD rats.The transmission electron microscopy was used for identifying the lutein cells by observation of the mitochondrias with the tubular cristae.Lutein cells were exposed to one of the following treatments:0,0.05,0.5,5,50 μg· mL-1 aconitine.Cell viability was determined by the MTT,and secretion of hormone was measured by RIA kit detection of estradiol and progesterone,and the effects of oxidative damage were measured by MDA and superoxide dismutase kit.RESULTS After 24 hours,compared with the control group,the high dose of aconitine(50 μg·mL-1)could inhibit the proliferation of lutein cells remarkably(P 0.01),secretion of progesterone was lower than that of the control group when the aconitine was at the levels of 0.5,5,50 μg·mL-1.CONCLUSION The results indicated that aconitine could induce the toxic effects on lutein cells above the level of 5 μg·mL-1,and aconitine can inhibit the proliferation of lutein cells and the secretion of hormone.
