摘　　要：目的建立HPLC测定苏木中巴西苏木素和原苏木素B含量的方法。方法色谱柱为Agilent Zorbax XDB C18(150mm×4.6mm,5μm),流动相为甲醇-0.1%醋酸(20:80),流速为1.0mL·min-1,检测波长为280nm。结果巴西苏木素和原苏木素B的线性范围均为0.025～0.50mg·mL-1,r均为0.9994;平均回收率分别为95.44%(RSD=2.57%)和94.69%(RSD=1.38%)。结论新建方法简单、快速、准确,适用于苏木中巴西苏木素和原苏木素B的含量测定。OBJECTIVE To develop an HPLC method to determine brazilin and protosappanin B in Caesalpinia sappan L.METHODS The Agilent Zorbax XDB C18（150 mm × 4.6 mm,5 μm）column were used with methanol-water（0.1% acetic acid）（20：80） as mobile phase.The flow rate was 1.0 mL·min-1 and the detective wavelength was 280 nm.RESULTS The peak areas and concentrations of brazilin and protosappanin B showed good correlation in the range of 0.025-0.50 mg·mL-1（r = 0.9994）.The average recoveries were 95.44% for brazilin with RSD of 2.57%,and 94.69% for protosappanin B with RSD of 1.38%,respectively.CONCLUSION The method is simple,suitable and accurate for the determination of brazilin and protosappanin B in Caesalpinia sappan L.
