摘  要:采用正交试验方法考察乙醇体积分数、提取温度、提取时间、料液比对苦苣菜总黄酮提取效果的影响,寻求苦苣菜总黄酮提取最佳工艺,并进一步建立总黄酮中芦丁含量的高效液相色谱(HPLC)法测定方法。结果表明,料液比对总黄酮提取量有显著影响,总黄酮提取量也受到提取温度和乙醇浓度的影响。最佳提取工艺:30%乙醇作为溶剂,提取温度80℃,料液比1∶20,提取时间2 h,此条件下总黄酮提取量达6.065 mg/mL。HPLC法测定黄酮中芦丁含量结果表明,在0.025-0.075 mg/mL范围内线性关系良好(R=0.999,n=5),平均回收率为97.98%,RSD为2.53%。HPLC法简便快捷、灵敏,结果准确,重现性好,可用于苦苣菜中芦丁含量的检测。The orthogonal design was used to study the effect of ethanol concentration,extract temperature,time and solid/liquid ratio on total flavonoids extract from Sonchus oleraceus.and establish a determination method of rutin content in total flavonoids by HPLC.The results showed that the solid/liquid ratio had the significant effect on total flavonoids quantity and the total flavonoids quantity was affected by extraction temperature and ethanol concentration also.6.065 mg/mL total flavonoids can be extracted under the optimum extract technological conditions of 30% ethanol,1∶20 solid/liquid ratio,at 80℃ for 2 h.The results of determining rutin content in total flavonoids by HPLC showed that there was a good linear relationship between peak area and rutin content with 0.025~0.075 mg/mL and its mean recovery and RSD were 97.98% and 2.53%,which indicates that HPLC with simplicity,sensitivity,accuracy and good repetition can be used in determination of rutin content in Sonchus oleraceus.
