摘 要:目的:观察独一味胶囊对大鼠实验性PTCA术后再狭窄的防治作用。方法:Wistar大鼠随机分为正常组、PTCA模型组、独一味胶囊(0.50、1.25、2.50g/kg)组,冠心丹参片(1.0g/kg,阳性对照组)。各组均于PTCA术前5天给药,末次给药2h后手术分离大鼠左颈总、颈外和颈内动脉。将2.0F球囊导管插入左颈总动脉,以l.5个大气压充盈球囊,来回拖动球囊3次以剥脱内皮,局部作用30min,建立颈动脉球囊损伤模型。于术后2周取损伤节段颈总动脉,进行病理检查及图像分析并测量内膜和中膜面积。结果:手术后2周大鼠颈总动脉损伤后引起血管内皮细胞剥脱,新生内膜形成并增生,导致管腔狭窄。经独一味胶囊治疗3周后,再狭窄大鼠动脉内膜面积、中膜面积及内膜/中膜面积比模型组显著减小,相对管腔面积增大,差异具有显著性。结论:藏药独一味胶囊可抑制内膜增生,对PTCA后的血管再狭窄具有防治作用。Objective:To observe the therapeutic effect of Lamiophlomis Rotata capsule on restenosis after experimental PTCA in the rat carotid injury model. Methods:Wistar rats were randomly divided into normal group,PTCA model group,model control group,Guanxin Danshen tablet group (1.0g/kg)and Lamiophlomis Rotata capsule group (respectively0.50,1.25,2.50g/kg Lamiophlomis Rotata capsule). All groups were administered 5 days before PTCA,the last administration in rats 2h after the surgical separation of the left common carotid,external carotid and internal carotid artery. The 2.0F balloon catheter was inserted in left common carotid artery,filling balloon to l.5 atmospheres,draging the balloon back and forth 3 times to endothelial denudation which was local role in 30min for establishment of carotid artery balloon injury model. Injuries to carotid artery segments were taken after 2 weeks,which were performed pathological examination and image analysis and measurement of intima and media area.Results:Vascular endothelial cell denudation,new intima hyperplasia and hyperplasia,resulting in lumen stenosis appeared in 2 weeks after carotid artery injury in rats. After 3 weeks treatment of the Lamiophlomis Rotata capsule,intimal area,medial area and intima/media area of restenosis in rat carotid artery were reduced significantly,and relative lumen area was increased significantly. Conclusion:The Lamiophlomis Rotata capsule could inhibit intimal hyperplasia,and play a positive role in the prevention of restenosis after PTCA.
