海南木莲基因组DNA提取及ISSR反应体系的优化

摘　　要：通过对不同提取方法获得的海南木莲DNA质量进行分析,发现采用预先去杂CTAB法提取的海南木莲DNA比其它方法简便、高效,DNA杂质少,得率高;然后通过单因素实验设计对海南木莲ISSR—PCR反应体系进行优化,确立了适于海南木莲的ISSR反应体系,即20μL反应体系中含40 ng模板DNA、0.15 mmol.L-1dNTPs、0.40μmol.L-1引物、2.5 mmol.L-1Mg2+和1.5 UTaqDNA聚合酶;最后用16个海南木莲样品对优化体系进行检验,结果表明该体系扩增效果稳定,特异性高,可以用于海南木莲分子生物学的进一步研究。The genomic DNA being extracted by predimpurity CTAB method is high quality with less impurity than other methods.By the single factor design of ISSR-PCR reaction system,a satisfactory ISSR reaction system was established.The concentration of the reaction system was DNA template 40ng,dNTPs 0.15mmol·L^-1,primer 0.40μmol·L^-1,Mg^2＋ 2.5mmol·L^-1,Taq DNA polymerase 1.5 U.The reaction system was tested with the samples of sixteen Manglietia hainanensis material.The pecial bands of amplication of ISSR-PCR were bright,orderly and are good to the following molecular marker analysis and biology research.
